This dataset contains the carbon chemistry measurements of a laboratory experiment study to understand the effects of ocean acidification on winter flounders. To address the goals of the NOAA Ocean Acidification Program (OA), the James J. Howard Laboratory constructed an OA experimental system to conduct OA experiments on commercially important species native to Atlantic waters of the NE U.S. The species of interest exhibit a broad range of ecological and life-history types from entirely marine to entirely estuarine. In order to conduct OA experiments on several different species, the required experimental system was able to mimic both estuarine and oceanic conditions. This system was designed to produce 5 pCO2 concentrations above an ambient pCO2 treatment and 5 temperature regimes. The system is also capable of manipulating dissolved oxygen, salinity, and other biogeochemicals common to a benthic habitat. The analytical laboratory conducts analysis according to the guidelines developed by the ocean acidification subcommittee of the Ocean Carbon and Biogeochemistry Program. When followed correctly, these guidelines should produce data with a high degree of accuracy and precision.
To construct a multi-variable ocean acidification experimental system fully costomizable to the target species for experimentation.

Parameter or Variable: Dissolved Inorganic Carbon;
Abbreviation: DIC;
Unit: umol/kg;
Observation type: Dissolved inorganic carbon;
In-situ / Manipulation / Response variable: Manipulation condition;
Measured or calculated: Measured;
Sampling instrument: manual;
Analyzing instrument: NDIR based DIC analyzer (Apollo SciTech Inc);
Detailed sampling and analyzing information: Column samples were collected by hand using a tube attached to the bypass at the base of each CO2 addition column. Experimental container samples were extracted and transfered to sample bottles using a 60 mL syringe. All samples were collected in a 300mL glass BOD bottle by filling from the bottom of the bottle. A 0.75 mL volume of each sample was analyzed on an Apollo SciTech DIC analyzer.;
Replicate information: no field replicates collected;
Standardization description: Different volumes of a CRM were analyzed and the area outputwas used to produce a standard curve. A CRM was also analyzed at the beginning and end of a daily sample analysis. The standard curve was used to determine the measured CRM and sample values from the coulometer output. The difference in the measured CRM value from its real value was used to determine measurement uncertainty.;
Standardization frequency: Daily;
CRM manufacturer: Dickson Lab at UCSD;
Preservation method: Saturated solution of mercuric chloride;
Preservative volume: 0.06mL;
Preservative correction: none;

Detailed sampling and analyzing information: Samples were collected according to location. Samples from the columns were collected in a 250mL plastic bottle and transfer by pouring into a 100mm spec cell. Samples from experimental containers or from DIC sample bottles were collected and transferred to a spec cell using a 60mL syringe. The spectrophotometer was then used to determine absorbances at wavelengths of 434, 578, and 730 for a sample without and then with 2mM m-cresol purple dye added. The absorbance values were used to determine pH according to the citation given below;

Non-dispersive infrared (NDIR) dissolved inorganic carbon (DIC) analyzer
A known amount of seawater is dispensed into a stripping chamber where it is acidified and purged with an inert gas. The amount of CO2 in the resulting gas stream is determined with a non-dispersive infrared (NDIR) carbon dioxide analyzer.

spectrophotometer
spectrophotometer, optical spectrometer, spectrograph or spectroscope
An optical spectrometer (spectrophotometer, spectrograph or spectroscope) is an instrument used to measure properties of light over a specific portion of the electromagnetic spectrum, typically used in spectroscopic analysis to identify materials.
Butler, L. R. P.; Laqua, K. (1995). "Nomenclature, symbols, units and their usage in spectrochemical analysis-IX. Instrumentation for the spectral dispersion and isolation of optical radiation (IUPAC Recommendations 1995)". Pure Appl. Chem. IUPAC. 67 (10): 1725–1744. doi:10.1351/pac199567101725. A spectrometer is the general term for describing a combination of spectral apparatus with one or more detectors to measure the intensity of one or more spectral bands."
